For wet sample loading, silica gel G is first swollen with eluent and then loaded into the silica gel column, and then the dissolved sample is poured into the silica gel column for separation. There is little difference in the substance separation effect between these two methods. The wet sample loading operation is simple and convenient, but less polar solvents such as methylene chloride and ethyl acetate are used to dissolve the sample. For some substances with low solubility, it is advisable to use the dry method for sample loading.
