Chromatographic column is the main component of high performance liquid chromatograph, and the column efficiency directly affects the analysis and detection of target compounds. However, in HPLC analysis, it is easy to cause problems such as high system pressure due to column blockage, peak tailing and widening due to low column efficiency, ghost peak due to column pollution and damage, etc. The internal causes of these problems are the dead adsorption of silanol, the presence of heavy metals, carbon loss, salt precipitation in buffer solution, and the drying of chromatographic column.


1、 Dead adsorption of silicon hydroxyl:


Due to the presence of silica gel hydroxyl groups on the surface of silica gel particles, the sample components are distributed between the mobile phase and the stationary phase, as well as between the mobile phase and the silica hydroxyl groups, that is, there is a double distribution effect. The adsorption of sample components by silicon hydroxyl is called dead adsorption or non-specific adsorption. When the adsorption of the sample components by the silanol group approaches saturation, the column efficiency decreases, which will lead to the tailing and broadening of the peak.


2、 Presence of heavy metals:


No matter how high the purity of silica gel particles as the base material of chromatographic column is, no matter how treated, heavy metals will remain on the surface of silica gel particles in the form of metal oxides. These metal oxides are easy to form chelates with other compounds, so that they can be oxidized to produce asymmetric peaks or tailing peaks.


3、 Carbon loss:


After long-term use of the stationary phase, some carbon chains will be eluted by the mobile phase and flow out of the chromatographic column with the mobile phase, causing carbon loss.


4、 Precipitation of salt in buffer solution:


During chromatographic analysis, the mobile phase sometimes contains buffer salt solution. After the analysis, if the aqueous mobile phase with a certain proportion is not used for flushing first, but the pure organic phase is directly used for flushing, the organic phase in the micro environment of the column is high and the aqueous phase is low at the moment. At this time, the buffer salt solution in the mobile phase is easy to precipitate salt, which blocks the column, increasing the column pressure and decreasing the column efficiency.


5、 Column drying:


If the chromatographic column is not properly stored, the preservation solution in the chromatographic column will volatilize completely and the column interior will become dry, which will damage the chromatographic column and affect the separation effect.
